(2) Postdoctoral position: efficient design of chromatin loops and recruitment of long non-coding RNAs (lncRNAs)
The three-dimensional structure of eukaryotic genomes plays a central role in gene regulation, and it is now recognised that abnormal structures can affect a wide range of diseases. However, few methods exist to correct the aberrant recycling and positioning of chromatin within the nucleus. We seek to develop methods to efficiently design chromatin loops and recruit long non-coding RNAs (lncRNAs) to move specific chromatin regions within the nucleus using a CRISPR-Cas system that targets genomic DNA or RNA. these methods will be used to regulate the expression of disease-associated genes on the X chromosome, which will enable an understanding of the role of spatial organisation in controlling gene expression and X chromosome inactivation, which will enable fundamental studies on the role of spatial organisation in controlling gene expression and X-chromosome inactivation. It will also provide new approaches for the treatment of diseases caused by X-linked gene mutations, allowing for precise restoration of the original expression levels of dose-sensitive genes. The proposed approach will utilise the X chromosome as a model system, but will also be applicable to genes in other regions of the genome.
