Scholarship Description:
The Xu laboratory studies the molecular and cellular mechanisms of human brain development and the pathogenesis of neurological disorders, focusing on human neurons, microglia and neuro-immune interactions using human induced pluripotent stem cell (hiPSC) models. Our research employs multidisciplinary approaches including stem cell reprogramming, molecular and cell biology, CRISPR/Cas9 gene editing, bulk and single cell RNA sequencing, patch clamp recording, pharmacology, animal behaviour and various imaging techniques. Our recent studies have successfully generated a hiPSC-based disease modelling system, including in vitro brain organoids and in vivo human-mouse chimeric brain models, to interrogate human neuronal and microglial function in brain development and neurological disorders, and identified potential therapeutic targets.
